Recognizes a cluster of proteins between 70-80 kDa, identified as tyrosinase. Occasionally a minor band at 55 kDa is also detected. This MAb shows no cross-reaction with MAGE-1 and tyrosinase-related protein 1, TRP-1/gp75. Tyrosinase is a copper-containing metalloglycoprotein that catalyzes several steps in the melanin pigment biosynthetic pathway; the hydroxylation of tyrosine to L-3,4-dihydroxy-phenylalanine (dopa), and the subsequent oxidation of dopa to dopaquinone. Mutations of the tyrosinase gene occur in various forms of albinism. Tyrosinase is one of the targets for cytotoxic T-cell recognition in melanoma patients. Staining of melanomas with this MAb shows tyrosinase in melanotic as well as amelanotic variants. CD106 (also known as vascular cell adhesion molecule-1, or VCAM-1) is a member of the Ig superfamily of adhesion molecules. It is expressed at high levels on cytokine stimulated vascular endothelial cells, and at minimal levels on un-stimulated endothelial cells. It is also present on follicular and inter-follicular dendritic cells of lymph nodes, myoblasts, and some macrophages. CD106 serves as a ligand for leukocyte beta-1 integrin VLA-4 and mediates cell adhesion of leukocytes to activated endothelium. They recognize pathogen-associated molecular patterns that are expressed on infectious agents, and mediate the production of cytokines necessary for the development of effective immunity. Fibronectin is a soluble dimeric glycoprotein of 440 kDa, which is present in cells, extracellular matrix, and blood. This MAb reacts with the cellular as well as plasma form of fibronectin. Reportedly, after iv administration, this MAb localizes to tumor vessels where it binds to the underlying basement. The epitope recognized by this antibody is not accessible to the circulating MAb in normal tissues, indicating that it can be used to specifically target tumor vessels in vivo.
